I use prime in my aged water so that i can get it ready for my daily wc... i do about 40% wc.. do i need to put more prime in when i do the wc from age water to main tank??

no need. but i would put prime in main tank instead aged water barrel. what you 're doing is ok for daily water change, but if you skip 1 day and let it sit over 24hrs, your water will be cloudy. Just my experience.

I also do not use prime in the aging barrel ~ it coats it and becomes somewhat slimey over time. Just the chlorinated water from the city keeps it very squeaky clean. Then, I just put prime in the tanks prior to adding water. I know many hobbyists on this forum do the same and I've had no issues doing it this way many years. Good luck.

I add prime to my tank and pump water straight from my tap to the tank.

No
All you need to do is add prime to the container holding your new aged water, Your water barrel.

If you were to add it to the tank you would have to use twice the amount because you must dose for the entire amount of water, aging barrel + what's already in the fish tank.
