you don't want it to be EXACTLY.. why not add 1" to all sides.. or 1/2".. so you have room for error.. you could make it larger.. its up to you
i am getting a 100 gallon tank and need help on deminsions for the stand i will be making....
the demensions for the stand are 48Lx24Hx20W... should i make this stand accomidating exactly to the tank 48x20?
also would like to add some type of molding to three sides (front left and right) so the tank fits in almost like a puzzle peice

it all depends on what you plan on putting inside the stand. In my recent build, i have 7/8-1" sticking out on all sides so that I could fit a 25G high tank inside the stand. Used as sump.

I always leave 1/4-1/2 extra on all sides. And try to remember 2x4's are Not 2x4 lol.
